The series revolves around Emily Elizabeth Howard, a young girl who owns Clifford, a big red dog who can speak to her for the first time since the late 1980s-early 1990s direct-to-video series.<ref name=people/> Together the two friends explore their island home, Birdwell Island, and embark on adventures.

Jetta, Vaz, Charlie, T-Bone, Mac, and Cleo are all removed from the reboot, with Samantha, Jack, Pablo, Tucker and Bailey replacing all the classic characters. The Speckle Story was also removed in Season 3, and the show went straight to the next segment. The Clifford’s Big Idea segments are replaced with Willa moments.

Samantha's two mothers caused the series to be controversial among anti-LGBT crowds.
